How do I test the package on MacOs and Windows?

Not tested on Mac OS or Windows: don't know how to because I don't have access especially to Windows system. Didn't hear good suggestions from Slack discussion.

This has been answered in several places, you can use r-hub to test the package, or use the bioconductor docker images, or use free continuous integrations tests to check your package on these OS. (Or all of the above)
