I recently gave a Training and received some user feedback on usability (and as far as I remember, this was a topic for almost all trainings):

In general it was about scaling/calibrating in vitro results for Peff (via CACO2) for intestinal permeability or intrinsic clearance rates for CYPs etc.

Even though PKSim provides all the infrastructure to implement measured in vitro values, it is sometimes hard to understand how my in vitro value should be (if at all) scaled to be entered for the different calculation methods I can choose from (in vitro in presence of microsomes or per g liver biopsy, ...). An additional level of complexity is added when loading relative expression values and defining reference concentrations.

So, is there someone in this community with a strong IVIVE focus and also using PKSim who would like to generate / provide a workflow guide (and maybe even a set of exercises for self-training) for this? ;-)

Hello,

I would love to help out with creating IVIVE workflows. I have done some recent work on IVIVE of hepatic clearance and overall have good working knowledge of this topic. I will look more into what you are looking for and will get back to you.

thanks,
Krina

@krinaj This is some great news.
My suggestion would be that you write down the info that you want to share in a wiki format (for example in your own repository using markdown). Your wiki could be for example integrated in the OSP wikis once finalized.

Thanks, Michael. I can write up some explanation of IVIVE of hepatic clearance with example and references in github repository wiki page.

Hello,

I have created this draft content on this topic. Can you please review and let me know of your comments?

I also have a question on slide 7. Can you please check if you have any insights in that regard?

Thanks,
Krina
