Hi, I'm working on a model for a CNS drug. Several questions bother me. Could anyone help me?

  1. What's the unit of brain tissue in the software? The Y-axis is ng/mL but normally the tissue concentration should be normalized with ng.
  2. In the simulation, the drug shows in brain tissue and interstitial but absolutely zero in intracellular. Is this regular? What's the difference between interstitial and intracellular concentration?
  3. I don't know the transporters for this drug, but I got the brain/plasma concentration ratio from animal PK study. Could I set the brain/plasma ratio fixed in the software?
