Summary

HTDoseResponseCurve is an R library that makes it straightforward to analyze and 
plot drug exposure experiments performed by high-throughput analysis of cells in 
6, 24, 96, or 384 well plates. This library is particularly suited to 
experiments where one or more cell plates have been measured repeatedly over 
time. Although HTDoseResponseCurve can read data from any source, it can 
natively consume the raw files exported by the [Incucyte Cell Analysis System](http://www.essenbioscience.com/essen-products/incucyte/).

*HTDoseResponseCurve* fits curves using the 
[drc library](https://cran.r-project.org/web/packages/drc), available on CRAN.

HTDoseResponseCurve grew out of our laboratory's need to fit dose response 
curves to drug screens in cell lines. These screens are typically performed on 
one or more 96- or 384-well plates, and may involve repeated capture of data 
about the cellular phenotype over time. 

Key Features

* Fits and plots dose-response curves from high-throughput experiments
* Reads data exported from the Incucyte platform (Essen Bioscience) in the 
Excel/XML format produced by that instrument. 
* Calculates standard descriptive values
    * SF50
    * Area Under the Curve (AUC)
    * P value for ANOVA testing difference in curve fits
* Calculates differential AUC 
    * for multiple treatment concentrations at a single timepoint
    * for a single treatment concentration over multiple timepoints
* Identifies time point at which AUC/SF50 is maximized
* Calculates Combination Index and Bliss synergy analysis

HTDoseResponseCurve does not perform image analysis.

HTDoseResponseCurve requires installation of three packages available on CRAN:

* [drc](https://cran.r-project.org/web/packages/drc)
* [readxl](https://cran.r-project.org/web/packages/readxl)

The *drc* package requires 
[multcomp](https://cran.r-project.org/web/packages/multcomp), which currently 
requires version 2.39-4 of 
[survival](https://cran.r-project.org/web/packages/survival). You may need to 
obtain this version from source, as *install.packages()* installs version 
2.39-3.
