It'd be nice to have a transparent pen, e.g. for coloring or highlighting. Maybe this is a good icon.

  • Toggle on/off with default 50%
  • When on, bring up a few more buttons for additional transparency amounts (e.g. 25% and 75%, or more). When toggled off, those buttons disappear. This lets you configure transparency, but without cluttering the interface when you're not using it.
  • Shift-click to toggle fill transparency separate from stroke transparency. I guess we need a 100% opacity option then, so one part can be 100% and another can be semitransparent.
  • Popup configuration for arbitrary opacity
  • Should opacity be global to the stroke, or adjustable via pressure? Maybe this is a configuration toggle.

does it have to be a boolean situation....

What about you press the button and it opens a slider (like thing) for 100 to 0 for opacity?
