Agenda

  1. Update on Atomwise. Approval secured from Diamond, and now awaiting approval from Warwick, before shipment of Atomwise compounds currently at UCL.

  2. Update on AZ compounds:
    i) Securing compounds from AZ (ongoing with Bart, Joe and Chris) and elsewhere (e.g. synthesis in Synthetic Design of Pyrazolopyrimidine #38 and shown below - first the AZ alcohol and then the amine derivative).
    ii) Plan for what to do with these compounds

  3. Evaluation of existing fragments in Diamond and Warwick, and plan for next fragment variants or screens.

  4. Aim remains:
    i) Structures of two mur ligases bound to the same small molecule
    ii) Identify new starting points for inhibitors

  5. Funding
    @drc007 identified and suggested this MRC opportunity, deadline May 12th. This is a possible scheme, though Chris Dowson said that the aims need to be broad (which potentially the open source approach can claim to be, in terms of community engagement). Likely that national funding schemes will be needed, i.e. separate UK and US schemes.

  • @mattodd to post meeting recording
  • @WieselHenson to synthesise the AZ alcohol compound and plan for the synthesis of the amine derivative
  • Chris Dowson to secure approval from Warwick to ship Atomwise compounds, then @danaklug to ship compounds to Warwick and Diamond.
  • @Rebecca-Steventon to screen @LizbeK SGC fragments and then @danaklug elaborated fragments
  • Laura setting up crystallisation trials in Warwick
  • Joe, Chris and Bart to pursue AZ compound shipment for crystallisation trials.
  • Chris Downon and Mat to consider the MRC scheme for funding.
