YouScope R2022 Beta 1
This is the first beta of the R2022 (v2.2.0) release of YouScope.
New functionality/changes:
-
New Nemesys Pump driver that supports automatic switching/refilling once a syringe is empty from / to a reservoir.
-
New imaging job to allow for the division of two out of focus frames for flat-fielding and high contrast membrane patterns for better cell segmentation (i.e. with CellX).
-
Includes latest device supporting new devices such as, but not limited to Nikon Ti2, Lumencor Light Engine.
Which YouScope version to choose
We only provide the Windows 64bit version of YouScope (YouScope_64bit_Installer.exe). Typically, one will choose the 64bit version of YouScope on Windows 64bit, except if one depends on drivers only available in 32bit (see previous releases). For legacy systems please use a previous release that still works perfectly fine but might lack novel features and new device drivers.
Measurement Viewer
It is possible to execute the Measurement Viewer as an own process, e.g. to analyze a measurement on a personal computer not connected to the microscope. The Measurement Viewer is available either for Windows 32 and 64bit (measurement-viewer.zip), or for Posix (MacOSX, Linux,...) systems (measurement-viewer.tar.gz).
Drivers
This version of YouScope is based on the drivers from Micro-Manager 2.0.1, nightly build from 21-April-2022 21:36. Revision/Commit/Sha-1: 6fc61ab30e4daf75c0bd2f782c2730a5a5ead538
Requirements
The Java Runtime version 1.7 or higher has to be installed ( http://www.oracle.com/technetwork/java/javase/downloads/jre8-downloads-2133155.html ). Note, that you need to install a 32bit Java Runtime if you want to execute YouScope in 32bit, and a 64bit Runtime if you want to execute YouScope in 64bit.
For the Nikon Ti2 you need to install the TIControl 2.x and SDK an copy the Ti2_Mic_Driver.dll
to the drivers64
folder of YouScope. The dll is located under C:\Program Files\Nikon\Ti2-SDK\bin
.
Latest Development Builds
A list of the 30 last development/nightly builds of YouScope. Don't expect these nightly builds to be stable, and rather use regular builds whenever possible!
YouScope R2018
Finally, after some time, we release YouScope R2018 (originally planned as R2017). As compared to the last stable release, the following features were added:
- Wells in a microplate measurement can now be grouped into different wells, and for each group a different imaging protocol can be defined.
- The focus score can be calculated for the images in the LiveStream.
- Jython support for scripting.
- Advanced Live cell detection via CellX
- HTML export now contains possibility to few measurement images, similar to the measurement viewer (currently only IE, Edge and Mozilla when using tiff).
- Measurements can now be paused and resumed. While paused, the microscope becomes unlocked and can be either controlled directly, or other measurements can be run. A paused measurement can, however, not be edited.
- Measurements now possess metadata and a free form description. Both is exported as XML and HTML, replacing the old scope_settings.xml.
- File names now allow for any special characters allowed by Windows, and not only a subset thereof.
Besides these new features, several old ones were improved and bugs were fixed.
Which YouScope version to choose
You can run the 32bit version of YouScope (YouScope_32bit_Installer.exe) both on 32bit and 64bit versions of Windows. Only on Windows 64bit you can run the 64bit version of YouScope (YouScope_64bit_Installer.exe). Typically, one will choose the 64bit version of YouScope on Windows 64bit, except if one depends on drivers only available in 32bit. It is also possible to download a combined 32bit/64bit version (YouScope_3264bit_Installer.exe), and dynamically switch between them without loosing any configuration settings.

Drivers
This version of YouScope is based on the drivers from Micro-Manager 1.4.22, SVN trunk r15693, 3rdparty r176, device interface version 65.
Some people reported that, for them, the drivers from Micro-Manager 1.4.20, SVN trunk r14990, 3rdparty r173,
device interface version 61 worked better (while others prefer the newer version). As a compromise, we provide an optional installer to "downgrade" the drivers to 1.4.20 (for win32, 64 and combined). This installer usually don't has to be executed, except if you experience problems with the new drivers. To downgrade, first normally install YouScope, and then run the downgrade installer on the same folder.

YouScope R2017 Beta 1
General Information
This is the first beta of the R2017 (v2.1.0) release of YouScope.
Since a lot of the code base considering how a measurement is run (new pause option) has changed, this version might not be as stable as the R2016 final release. Choose R2016 final release if stability is more important for you than the new features offered by this version.
New functionality/changes:
- Measurements can now be paused and resumed. While paused, the microscope becomes unlocked and can be either controlled directly, or other measurements can be run. A paused measurement can, however, not be edited.

YouScope R2016
General Information
Finally, this is the release of R2016 (v2.0.0) of YouScope.
Only little was changed as compared to Beta 6, for which we did not receive any error reports. The only major changes was adding new tooltips for jobs and measurements (i.e. providing some more in program help), and adding the binaries for the Nemesys driver (thanks to Cetoni), which are now also registered automatically.
This version is stable and ready for productive use!
YouScope R2016 Beta 6
General Information
This is the sixth beta of the R2016 (v2.0.0) release of YouScope.
Most of the changes in this version consider the support of non-rectangular microplates. Since a lot of the code base considering microplate measurements had to be changed, this version might not be as stable as the other beta releases. Choose Beta 5 or a later version if stability is most important for you.
New functionality/changes:
- Support of custom, non-rectangular microplates. The microplates can have virtually any layout, including gaps, missing wells, different well sizes, and unregular positions.
- Path optimizers were revisited: there are a few more options in the different optimizers, e.g. different metrics.
- Complete refactoring of the microplate measurement: Position configuration should be significantly simpler, and configured positions can manually be changed afterwards.
The Measurement Viewer is available either for Windows 32 and 64bit (measurement-viewer.zip), or for Posix (MacOSX, Linux,...) systems (measurement-viewer.tar.gz).
YouScope R2016 Beta 5
General Information
This is the fifth beta of the R2016 (v2.0.0) release of YouScope.
A major bug, present since the last release, concerning possible data loss when imaging microplates was fixed. In this bug, the image file names did not contain an identifier of the y-position of the well. Imaging at different y-positions thus could have led to overwriting of files. The bug is only present in R2016 Beta 4. Beta 3 or lower do not include this bug.
New functionality/changes:
- New path optimizers for microplate measurements. The path can now be optimized using either a 1.5- or a 2-approximation to the traveling salesman problem, which typically return very good solutions even for very irregular positions. Furthermore, the path can be visualized.
- Two new jobs, considering a new way to stagger and precise waiting times between executions of a given job, were added.
