Will it be possible to use a Let's Encrypt certificate to sign client certificates yourself, or will it be possible to request client certificates from Let's Encrypt?

Not on the roadmap for Let's Encrypt. Generally my understanding is that if you'd like to issue client certificates for your service, you sign them using a private key that you hold, and validate them with the same private key. There's not typically a need to involve a public CA.
