hello! I have a problem with using SOAPtyping... I know the HLA type of my sample (HLA-A02:01 and HLA-A25:01) and i just wanted to check the PCR kit I received. I have already read the "tutorial" but it's not enoughto succeed in my analysis.
As you can see on the picture, I have a lot of mismatches (blue, some red and some green), especially in the exon 2... In the datasheet of the PCR kit, it is explained that red and green mismatches must be solved manually... But, if we look at the green one in exon 3, there is no mismatch with the right HLA alleles... I really don't understand.
Honestly, I am confused, I spent all morning on this and I am quite worried about using Soaptyping for an unknown HLA sample ...
