Masternode created with desktop app together with owner's address and operator address. However, the masternode will not run when i tried to enable it and have to go remove gem=1 in the defi.conf file.

The procedure is as follows, whether Nodehub, Allnodes or mydefichain:
You get a legacy address as OPERATORADDRESS from your provider (Allnodes) and create your own legacy address in your local wallet (Owneraddress).
Then the command in the CLI: createmasternode OWNERADDRESS OPERATORADDRESS
Wait 8h until the Masternode is ENABLED and you are done.
DO NOT click on ENABLE in the wallet. Just leave it as it is. Do not enter gen=1 or anything else in the local config, it is not necessary. Close wallet, store PC/Laptop in the safe - done :-)

Thankyou I will give it a try. I have just try to resign the wallet first. So I will have to wait for the resign to be finish and try to runthe masternode again right?

Yes, but you need a new Operator and new Owneraddress to create a masternode again.

Hello, I still cannot see my nodes minting any blocks do you have any suggestions?

Average Time to find a Block is ~4,5 days. It can take longer, maybe 14 days for your First Block.
