Supporting matfile function for very big data

Thank you again for your great work.

I want to use your code for very big data, a mat file that is more than 25 GB. And it's really difficult to load all the data on RAM and use your code. I am wondering if your code can support matfile function (http://www.mathworks.com/help/matlab/ref/matfile.html)
or make a solution for huge data that is not possible to load on RAM.
