In Illustrator

Create the path shape you want.
Add a rectangular path on a layer below it.
Cut the path shape with Cmd + X and add it as a top layer with Cmd + F.
Select both layers.
Click subtract in the Path Finder.
Re-add the original layer (currently in the clipboard) using Cmd + F.
