Can I join you guys tomorrow, so I know how to prepare these too? I'm available before 9:45am, and between ~11:30 and 1:20pm.

Also, before these get sent out I'd like to add a dot of histology ink to one oyster in each cassette for orientation. Also, I have 4 more cassettes to add to the pile; I just need to move them to the stabilizer tomorrow morning.

I'm not sure if there's any preparation. Aren't these ready to go? To my knowledge, I just need to verify if they need to be shipped as "dangerous goods in excepted quantities". If yes, then I have to do the packaging/shipping, as it requires university training to ship those types of items. If not, then there's not much else; just box 'em up and send 'em off...

Regardless, I'll meet with Grace when she comes in and see what's up. I have to help someone in the Friedman Lab for a bit at 11:30, so won't really be available then (not sure how long it's going to take). Maybe you can hook up with Grace when you're available at that time to see what's up?

OK, looks like the stuff is hazardous goods, so I'll have to do the packaging/shipping. @laurahspencer - fill out this form and print it out so I can include it with the package.

Here's info needed for the sheet:

  • Leave "PO Number" blank.

  • The first three columns are to indicate the labels on your cassettes. If you don't have some sort of sub-numbering system (e.g. Sample 1-1, Sample 1-2; where Sample 1 could go in the "Accession#" column), you'll have to list out each cassette's label on an individual line.

  • Type of specimen - Self explanatory

  • H&E - This is the desired stain. Mark with an X or check mark

  • Special instructions - Write "down is down"

I've prepped stuff so that it's compliant for shipping hazardous goods in excepted quantities. @grace-ac has all the shipping stuff needed and will be ready to go when you have the samples and sample sheet finished.

Ok, sounds good. @yaaminiv and I need to confirm that all the labels are legible and make sense; I'll let you know when that's done.

Where are the cassettes currently?

A few of things:

  • What's the end use of these samples (i.e. slides or RNA/DNA/protein isolation)?

  • If these are just being used for slides, then why did we use the PAXgene Stabilizer Kit?

  • If these are eventually going to be used for RNA/DNA/protein (this would be the reason to use the PAXgene reagents), then these samples should be stored in a refrigerator (up to 7 days storage at room temp) to prevent degradation of RNA/DNA/proteins...

I'm concerned that there hasn't been a response to @sr320 's and my last posts.

@yaaminiv and @laurahspencer and @grace-ac : Have these cassettes been moved to different storage conditions? If yes, please post here confirming they have been moved and please provide the location. If not, please move them, and then post here confirming they have been moved and please provide the location.

I don't know how strict the storage guidelines are, but this is well past the 7 days at room temp that's recommended by the manufacturer to preserve RNA/DNA/proteins in these samples...

Sorry, I dropped the ball on addressing this issue - I moved the cassettes into the refrigerator on Friday, in response to your comment; Yaamini & I need to check them out ASAP (this afternoon or tomorrow) to see if the labeling is clear, and to fill out that histology form.

Also - we're in class now, but afterwards we'll come check in with you to confirm that the location in the refrigerator is OK - @kubu4 will you still be around at 4pm?

@kubu4 the samples are currently stored in the white refrigerator in 209, bottom right corner in two white boxes:

histo boxes

And I left the histology processing record sheet on your keyboard; they should be all ready to go. If I can do anything to help get them packaged up let me know - I'll be around on Thursday mid-day.

@kubu4 I'm so sorry! I didn't see this message until just now. But I have the shipping forms still. I can leave them in lab if you want to send them out Monday. Or I can send them out Monday no problem.

I hadn't grabbed a box yet, but we have the absorbent material and large plastic bags.
