Minimum requirements for identification of a "reference matched" feature

Hi,

I have one specific question to the character of "reference matched" features.
Is - in theory - the spectrum from one single sample sufficient to finally annotate a feature as “reference matched” or is it mandatory to have several (how much?) identical hits from different samples?
