Natural Generation

Peyote grows wild in Sandy or Mountain biomes that are also Hot, with roughly a 4% chance per chunk to spawn (it needs both matching biome tags, so a hot desert or a hot mountain specifically). Unlike other crops, it generates as a single already fully-grown plant rather than a patch.

Crop generation can be toggled off via the Config, so if you can't find any, it may be disabled.

There's no way to buy Peyote from a Herbalist Villager - you'll need to find some growing naturally.

Farming

Peyote doesn't need a separate seed item - just place the Peyote item directly. It can go on any solid block, and doesn't need farmland or a particular light level to grow.

Instead of growing upwards like other crops, Peyote just ages in place - each stage it grows, it drops one more copy of itself when harvested, so breaking a fully-grown plant gives 4 Peyote. Once fully grown it stops ageing, and instead has a chance to spread, sprouting a new Peyote plant somewhere nearby. Bonemeal speeds up both ageing and spreading.

Crafting

Craft a Peyote Joint with 2 Paper and 1 Dried Peyote.

Fluid Crafting

Peyote Juice can be crafted in a Vat, 7 Dried Peyote with 4000mb of Water gives 4000mb of Peyote Juice.
