Exoglycosidases Mode of Action

Commonly, used exoglycosidases for glycan structure characterisation

  • ABS: Arthrobacter ureafaciens sialidase releases α(2-6/3/8)-linked nonreducing terminal N-acetylneuraminic acid (NANA, Neu5Ac) and N-glycolylneuraminic acids (NGNA, Neu5Gc).
  • NAN1: Streptococcus pneumoniae sialidase releases α(2-3)-linked nonreducing terminal sialic acids (NANA and NGNA).
  • BTG: bovine testes β-galactosidase hydrolyzes nonreducing terminal galactose with β(1-3/4) linkages.
  • SPG: S. pneumoniae β-galactosidase hydrolyzes nonreducing terminal galactose β(1-4) linkages.
  • AMF: almond meal α-fucosidase releases α(1-3/4)-linked nonreducing terminal fucose residues except core α(1-6) fucose.
  • BKF: bovine kidney alpha-fucosidase releases α(1-2/6) fucose-linked nonreducing terminal fucose residues more efficiently than α(1-3/4)-linked fucose.
  • GUH: β-N-acetylglucosaminidase cloned from S. pneumoniae, expressed in Escherichia coli, will digest β(1-4)-linked GlcNAc to mannose but not a bisecting GlcNAc β(1-4) linked to mannose.
  • CBG: Jack Bean α(1-3,4,6)-Galactosidase Hydrolyses α(1-3,4,6)-linked terminal galactose residues.
  • JBM: Jack Bean α(1-2,3,6)-Mannosidase releases non-reducing terminal α(1-2,6)-linked mannose residues more efficiently than α(1-3).
