Use the following recipe to prepare TAP medium.
| Order # | Solution | Amount |
| 1 | ddH2O | 967 mL |
| 2 | 1M Tris base (e.g. Trizma) | 20 mL |
| 3 | Phosphate buffer II | 1.0 mL |
| 4 | Solution A | 10.0 mL |
| 5 | Hutner's trace element | 1.0 mL |
| 6 | Glacial acetic acid | 1.0 mL |

100 mL of Phosphate buffer II: Dissolve 10.8 g K2HPO4 + 5.6 g K2H2PO4 in ddH2O, then top up to 100 mL with ddH2O.

500 mL of Solution A: Dissolve 20 g NH4Cl +5 g MgSO4• 7H2O + 2.5 g CaCl2• 2H2O in ddH2O, then top up to 500 mL with ddH2O.
***Omit** NH4Cl** when preparing Solution A for nitrogen-free TAP (TAP-N)
- 500 mL of 1M Tris base: Dissolve 60.57 g Tris Base in ddH2O, then top up to 500 mL with ddH2O.
If you do not add the solution components of TAP in order, your TAP medium might not work.
